Aging and genetic instability in yeast.

There is a striking link between increasing age and the incidence of cancer in humans. One of the hallmarks of cancer, genomic instability, has been observed in all types of organisms. In the yeast Saccharomyces cerevisiae, it was recently discovered that during the replicative lifespan, aging cells switch to a state of high genomic instability that persists until they die. In considering these...

Increased Genome Instability in Aging Yeast

In the September 26 issue of Science, McMurray and Gottschling (2003) report that aged yeast cells display high rates of loss of heterozygosity. Furthermore, they show that this reflects an impaired ability to correctly detect and repair DNA double-strand breaks. These results provide insights into how aging can engender genomic instability in eukaryotic cells.

Stochastic resonance with a single metastable state: Thermal instability in NbN superconducting stripline resonators

We study thermal instability in NbN superconducting stripline resonators. The system exhibits extreme nonlinearity near a bifurcation, which separates a monostable zone and an astable one. The lifetime of the metastable state, which is locally stable in the monostable zone, is measured near the bifurcation and the results are compared with a theory. Near bifurcation, where the lifetime becomes ...

A Microarray-Based Genetic Screen for Yeast Chronological Aging Factors

Model organisms have played an important role in the elucidation of multiple genes and cellular processes that regulate aging. In this study we utilized the budding yeast, Saccharomyces cerevisiae, in a large-scale screen for genes that function in the regulation of chronological lifespan, which is defined by the number of days that non-dividing cells remain viable. A pooled collection of viabl...

Genetic instability in budding and fission yeast—sources and mechanisms

Cells are constantly confronted with endogenous and exogenous factors that affect their genomes. Eons of evolution have allowed the cellular mechanisms responsible for preserving the genome to adjust for achieving contradictory objectives: to maintain the genome unchanged and to acquire mutations that allow adaptation to environmental changes. One evolutionary mechanism that has been refined fo...
